1) 1 + tan^2Θ = sec ^2Θ 
    So,  1  = sec ^2Θ - tan^2Θ 
    Ans : 1

1) sinΘ=4/5, Therefore Opposite=4, Hypotenuse=5,Adjacent=3

So, (cot^2x -sin^2x)/tan^2x = [(3/4)^2 -(4/5)^2]/(4/3)^2

                                                    =(9/16-16/25)/16/9
                                                    =(225-256/400)*9/16
                                                    = - 31*9/400*16
                                                    = -279/6400
The answer is bit weird but that's what it is. You should write these equations down on paper so that you can understand better. Happy Trigonometry time!!
